Determination of HLA-DRB1*04 Subtypes by Reverse dot-blot Hybridization and SSCP: Association between HLA-DRB1*04 Subtypes and Rheumatoid Arthritis in Koreans

Abstract
To determine which HLA antigens are associate with rheumatoid arthritis (RA), the HLA class strand comformation polymorphism (SSCP) in 105 rheumatoid arthritis (RA) and 235 healthy Koreans.
@ES The results were as follows:
@EN 1. The genotype frequency of HLA-DRBI*04 was 56.2% in RA patients and 29.8% in healthy normal controls (RR=3.0, p<10E-5), and the frequencies of the HLA-DRB1*15 (RR=0.5, p<0.05), and*14 (RR=0.4, p<0.05) were decrease in RA patients compared
with
healthy normal controls.
2. The frequencies of the HLA-DOA1*03 (RR=3.6, p<10E-5), DQB1*04 (RR=3.3, p<10E-3) alleles were increased and of the DQA1*01 (RR=0.4, p<10E-7), *o5(RR=0.5, p<0.05), DQB1*02 (RR=3.3, p<0.005), *06 (RR=0.6, p<0.01) alleles were decreased in RA
patients
compared with healthy normal controls.
3. The alleles of the HLA-DR4 subtypes (0401, 0403, 0404, 0405, 0406, 0407, 0410) were identified in Koreans, and the high frequent alleles were 0405 (11.1%), 0406 (6.4%), 0403 (5.1%) in healthy normal controls and 0405 (42.9%), 0406 (6.7%) in RA
patients. The frequency of the HLA-DRB1*0405 (RR=6.0, p<10E-6) was especially increased in RA patients compared with healthy normal controls.
4. HLA-DRB1*0405-DQA1*03 (HF=0.1876, p<10E-5) and HLA-DRB1*04 (HF=0.1160, p<10E7) haplotypes have significant positive linkage disequilibria. It suggests that HLA-DQA1*03 and DQB1*04 may react synergistically in the development of the RA along
with
HLA-DRB1*0405 alleles. These results showed that HLA-DRB1*0405 was strongly associated with rheumatoid arthritis in Koreans.
